Abstract


-Abstract-
To obtain species-specific DNA probe for detection and identification of
Camphyrobactor rectus ATCC 33238, 1) DNA extraction of Camphyrobactor rectus
ATCC 33238 and digested with the endonuclease Xba 1, 2) construction of the genomic
DNA library and screening of restriction fragment of genomic DNA, and 3) identification
of species-specific DNA probe by Slot blot hybridization method were done, Nine
restriction fragments of Camphyrobactor rectus ATCC 33238 genomic DNA were
obtained, Two of them, 1.8kbp (C2-15-1), and 0.6kbp (C2-15-2) were used by DNA
probe in this study. Slot blot analysis with restriction-digested genomic DNAs of
several periodontal pathogens, including three strains of Fusobacterium nucleatum, three
strain of Porphyromonas gingivalis, one strain of Prevotella intermedius, three strains of
Actinomyces gingivalis 381 £¦W50, Actinomyces actinomycetemcomitans ATCC 33384
and probe C2-15-2 was hybridized to Camphyrobacor rectus, Porphyromonas gingivalis
381 £¦ W50, Fusobacterium nucleatum ATCC 25586. These data indicate that further
study is need to obtain species-specific DNA probe for detection of C. rectus ATCC
33238.
